Comments: "We generated a soluble chimeric protein containing the extracellular carbohydrate recognition domain (CRD) and stalk region of the murine receptor, fused to the Fc portion of human IgG1 (Fig. 1B). The soluble Dectin-1 Fc chimera (FcDectin-1) was generated by cloning the CRD and stalk region of murine Dectin-1 into the pSecTag2 expression vector (Invitrogen Life Technologies, UK) containing the mutated Fc portion of human IgG1, described previously (Ettinger et al., 1996). The Dectin-1 CRD was amplified by PCR using the primers: 5′-aaaggtacctagcattttggcgacac-3′ (forward) and 5′-aaagaattccagttccttctcacagat-3′ (reverse). To generate soluble protein, the chimeric construct was transfected into HEK293T cells (IIDMM cell bank, UCT) using Lipofectamine (Roche Molecular Biochemicals, UK), according to the manufacturer's specifications."
