Blockages can be detected by the failure of the FCM to successfully plot data in the scatter plot even though the machine is in the ‘acquire sample’ step. This will be visualized on the scatter plot as either extreme alterations to the forward and side scatter plot (a Etch A Sketch like drawing) or a complete absence of cell populations that should be there otherwise. If blockages are not addressed immediately, counts will be significantly affected because the machine cannot tell when there is a blockage- it will think volume is passing through the flow cell when it isn’t, which will artificially lower your cell counts. 1 2017 Steven Wilhelm 2017.
