Comments: Immune sera against inclusion bodies were generated by the immunization of 6-week-old female NMRI mice (Charles River Laboratories) with 100 μg recombinant protein emulsified in Freund’s incomplete adjuvant (Sigma-Aldrich) followed by a boost after 4 weeks. Immune sera were collected 10 days after the second immunization via heart puncture. Comments: Anti-PfRh4 mAbs were produced at the Monoclonal Antibody Facility at the Walter and Eliza Hall Institute. BALB/c and C57Bl6 mice received three immunizations of recombinant PfRh4 purified as described below. At day 0, Complete Freund's adjuvant was mixed with the antigen into an emulsion and injected intraperitoneally. At day 30 and day 60 the antigen was mixed with incomplete Freund's adjuvant, and the emulsion was injected intraperitoneally. Serum ELISA titrations were performed at day 72. The mouse with the best response received a final injection of antigen in saline, and splenocytes were harvested 3 days later. Spleen cells were fused with SP2/0 myeloma cells to form B-cell-myeloma fused cells (hybridomas). Hybridomas were grown in hypoxanthine-aminopterin thymidine growth medium. ELISA was used to select hybridomas producing antibodies specific to PfRh4. Hybridomas were cloned by limiting dilution in multiwell plates aiming for one cell or less per well. The subcloning supernatants were screened by ELISA. Two or more rounds of limiting dilution cloning were generally required before the hybridomas were deemed monoclonal. The antibodies were purified from monoclonal hybridoma supernatants with protein A-Sepharose.

Can't find your Antibody?

We recommend that you click next to the search bar to check some helpful tips on searches and refine your search firstly. If you want to find a specific antibody, it's easier to enter an RRID or a Catalog Number to search. You can refine the search results using Facets on the left side of the search results page. If you are on the table view, you can also search in a specific column by clicking the column title and enter the keywords.

If you still could not find your antibody in the search results, please help us by registering it into the system — it's easy. Register it with The Antibody Registry (an Antibody Registry account is required. Create a free Antibody Registry account if you don't have one yet). An RRID will be generated in 1-2 business days.
