Summary: Keyhole limpet hemocyanin (KLH) is a cooper-containing protein comprising of subunits with MW of 400 kDa. This protein is found in the hemolymph of the sea mollusk Megathura crenulata. It has the ability to enhance the host’s immune response by interacting with monocytes, T cells and macrophages. KLH has been used primarily as a carrier for vaccines and antigens [1]. It was found that chicken immunized with KLH bound peptide raised an anti-KLH immunoresponse [2]. This can be tested by a single method such as the Ouchterlony technique.Reference1. Summary: This test shows the ability of Ab-3 antibody to inhibit the growth of S. aureus in vitro. The amounts of samples tested were 18 samples from chicks fed with hyper-immune eggs and therefore have the Ab-3, and six samples from chicks fed non-hyperimmune eggs and defenetively do not have the Ab-3. Inhibition of the growth of the bacteria plated out in blood agar was observed only when the Ab-3 was added to said culture media, sugesting that the anti-anti-anti-SpA (Ab-3) bound to the S. aureus and inhibited their ability to proliferate in culture. On the other hand, bacterial growth was observed when blanks or sera of chicks fed non-hyper-immune eggs were used because these samples lacked the protective Ab-3 [1]. Summary: Chemical synthesis facilitates the generation of peptides which are very difficult to express in bacteria, peptide/protein backbone modification, the incorporation of unnatural amino acids, and the production or synthesis of D-proteins. The C-terminal cysteine can be added to the amino acidic sequences of HIV peptides (fragment 579-601 of the HIV-gp41 and fragments 308-331 or 421-438 of the HIV-gp120). These peptide fragments were dimerized by cysteine oxidation with dimethyl-sulfoxide [1] to faciliutate their conjugation to keyhole limpet hemocyanin that acts as a carrier protein. Reference:1. They play essential roles in the activation and differentiation of immune cells, as well as proliferation, maturation, migration, and adhesion. They also have pro-inflammatory and anti-inflammatory properties. The primary function of interleukins is, therefore, to modulate growth, differentiation, and activation during inflammatory and immune responses. Interleukins consist of a large group of proteins that can elicit many reactions in cells and tissues by binding to high-affinity receptors in cell surfaces. Macrophages mostly make IL-18, which can be produced by hepatocytes and keratinocytes. Its principal target is a co-factor in Th1 cell induction. Summary: IgG is a monomer with an approximate molecular weight of 146 Kd and a serum concentration of 9.0 mg/mL. It is synthesized mostly in the secondary immune response to pathogens. IgG can activate the classical pathway of the complement system, and it also is highly protective. The four subclasses of IgG include IgG1, IgG2, IgG3, and IgG4. IgG crosses the placentae, protecting the neonate from infectious diseases. [1]
