Summary: We developed a supersensitive capillary reaction kit intended for the direct and qualitative detection of SARS-CoV-2 virus in saliva and nasal swabs. It enables sensitive and specific detection of the SARS-CoV-2 spike antigens in respiratory specimens more accurately than most PCR method within 5 minutes. Our rapid capillary kit will help not only fight but also end COVID-19 by providing the first sensitive self-diagnosis at home as well as POC clinical diagnosis at the health care facilities.

Summary: Barcoding or multiplexing is useful when the amount of data required per sample is less than the total amount of data that can be generated from a single flow cell: it allows a user to pool multiple samples and sequence them together, making more efficient use of the flow cell. The PCR-cDNA Barcoding Kit is used to prepare cDNA for nanopore sequencing for up to 12 samples, from an input of as low as 1 ng poly-A+ RNA. Users who do not have poly-A+ enriched RNA can use 50 ng of total RNA but additional optimisation may be required.Taking full-length poly-A+ RNA, complementary strand synthesis and strand switching are performed using kit-supplied oligonucleotides. The kit contains 12 primer pairs which are used to generate and then amplify double-stranded cDNA by PCR amplification: each primer pair contains a barcode and 5’ tag which facilitates the ligase-free attachment of Rapid Sequencing Adapters. Amplified and barcoded samples are then pooled together, and Rapid Sequencing Adapters are added to the pooled mix.

Authors: Rachel Aronoff, Guy Aidelberg
Group: XPRIZE Rapid Covid Testing, Guy Aidelberg
Summary: Corona Detective is based upon a molecular amplification strategy inspired by the ‘GMO Detective’ method, in order to detect the virus causing Covid-19. Done not only without complicated equipment, but with a simple +/- readout, the Corona Detective is very specific. Furthermore, controls to ensure sensitive detection, without false positives or negatives, are intrinsic to this solution. The final product, strips/plates of tubes with dry reagents, specific for Corona and a control gene (extraction control), can be shipped anywhere, without cold-chain dependence. Critically, monitoring tests could be run by ordinary people, by following this protocol. (Even kids have successfully participated in GMO detective workshops, also soldering their own fluor detectors.)This user guide will allow you to test samples with this system, provided as freeze-dried reaction components in 0.2ml tubes (formats possible, 8-tube strips, 96-well plates).Human or clinical samples should only be run in settings with access to appropriate biosafety facilities, of course.Acknowledging the JOGL Open Covid 19 Initiative and all the #proj nucleic-acid-amplification team for support.

Summary: FLASH amp is a method that allows for room temperature detection and amplification of a viral RNA sequence of interest submitted as part of the Xprize competition. The main components of the reaction are not included here and are given generic names such as enzyme X as they are currently not protected under IP. The steps outlined with tubes labeled with the generic names would allow any lab to reproduce the method. Version 1 of this method outlines the protocol for a fluorescence-based readout that can be used with a qPCR machine or TECAN style plate reader. Version 2 (to be released shortly) will be a colorimetric readout that can be assessed by eye. The outlined protocol assumes the user performing this in a laboratory setting will setup reactions in a clean room and analyze results in a seperate post-amplification room. In addition, since our amplification is rapid and at room temperature, care but be taken to add ligand to the master mix quickly and the reactions be sealed.

Can't find your Protocol?

We recommend that you click next to the search bar to check some helpful tips on searches and refine your search firstly. If you want to find a specific protocol and you know the DOI of the protocol already, it's easier to enter a DOI to search. You can refine the search results using Facets on the left side of the search results page. If you are on the table view, you can also search in a specific column by clicking the column title and enter the keywords.

If you still could not find your protocol in the search results, please help us by adding it into the system — it's easy. Create and publish your protocols at Protocols.io.
