Vector Backbone Description: Backbone Size:4456; Vector Backbone:pRS403; Vector Types:Yeast Expression, Integrating plasmid; Bacterial Resistance:Ampicillin
References:
Comments: GAL1 promoter, GFP silencing construct and CYC1 terminator fragment was cloned out of pYES2.1 and into pRS403.
The insert sequence can be found by clicking on the "View sequence" link. The rad9 intron loop is shown in upper case letters.

Species:
Genetic Insert: URA hairpin
Vector Backbone Description: Backbone Size:4456; Vector Backbone:pRS403; Vector Types:Yeast Expression, Integrating plasmid; Bacterial Resistance:Ampicillin
References:
Comments: 339bp of URA3 sequence from pRS406 was cloned into pYES2.1 GFP weak silencing construct in the sense orientation between PvuII and XhoI (replacing the GFP) and in antisense between EcoRI and XbaI sites. The expression cassette containing GAL1 promoter, URA3 silencing construct and CYC1 terminator was moved into pRS403.
The URA3 insert sequence can be found by clicking on the "View sequence" link. The rad9 intron loop is shown in upper case letters.
Please note that Addgene's sequencing results found a single nucleotide mismatch in the URA3 sequence at bp#3470 when compared to the full plasmid sequence provided by the depositing laboratory. This mismatch is not a concern for the function of the plasmid.

Genetic Insert:
Vector Backbone Description: Backbone Size:5857; Vector Backbone:pYES2.1 modified; Vector Types:Yeast Expression, Integrating plasmid; Bacterial Resistance:Ampicillin
References:
Comments: A MCS containing XhoI and EcoRI sites was cloned between the PvuII and XbaI sites of pYES2.1. The empty vector was created by replacing the hairpin of pIp-strongSC_GFP with XhoI and EagI sites
To make this plasmid integrating, the 2micron and f1 origins were replaced (using NheI and SpeI sites with sequence from S. castellii sc633:288301-289016 (amplified from genomic DNA with 5'-AAAAGCTAGCGATCCCTTATCAAATATGGTAC and 5'-AAAAACTAGTGTAGAATCCAGAGAATAGAATC).
Please note that Addgene's sequencing results differ from the full plasmid sequence from the depositing laboratory by a single nucleotide mismatch at bp# 3135 in the vector backbone. This mismatch is not known to affect plasmid function.

Can't find your Plasmid?
We recommend that you click next to the search bar to check some helpful tips on searches and refine your search firstly. If you want to find a specific plasmid, it's easier to enter an RRID or an Addgene Catalog Number to search. You can refine the search results using Facets on the left side of the search results page. If you are on the table view, you can also search in a specific column by clicking the column title and enter the keywords.
If you still could not find your plasmid in the search results, please help us by registering it into the system — it's easy. Register it with Addgene.
